【摘要】目的 研究葛根素联合阿托伐他汀对股骨头缺血性坏死Hedgehog信号通路的影响。方法 将50只SD大鼠随机分为5组,即空白对照组(Ⅰ)、模型组(Ⅱ)、阿托伐他汀组(Ⅲ)、葛根素组(Ⅳ)、联合用药组(Ⅴ),每组10只,Ⅱ、Ⅲ、Ⅳ、Ⅴ组均利用LPS联合甲强龙造大鼠激素性股骨头缺血性坏死模型,造模的同时Ⅲ组、Ⅳ组、Ⅴ组分别灌胃阿托伐他汀、葛根素、联合用药治疗,1次/d,连续8周后处死,采样待检。HE染色考察大鼠股骨头坏死情况;ELISA检测血清BGP、OPN、TGF-β1含量;RT-PCR检测股骨头组织Bcl-2、Osx、Runx2 mRNA表达量;WB检测股骨头组织VEGF、p53蛋白含量;免疫组化检测股骨头组织Smo、Gli蛋白含量;TUNNEL检测细胞凋亡。结果 较单一用药,联合用药组可明显提高血清BGP、TGF-β1含量、股骨头组织Osx、Runx2 mRNA表达及VEGF、p53蛋白含量(P<0.05),明显降低血清OPN含量、股骨头组织Bcl-2 mRNA表达及Smo、Gli蛋白含量(P<0.01),改善股骨头坏死及细胞凋亡情况。结论 葛根素联合阿托伐他汀能够通过影响Hedgehog信号通路,减轻股骨头的坏死程度。 |

Effect of Puerarin combined with Atorvastatin on Hedgehog signaling pathway in avascular necrosis of the femoral head |

[Abstract] Objective To study the effect of puerarin combined with atorvastatin on Hedgehog signaling pathway in avascular necrosis of the femoral head. Methods Fifty SD rats were randomly divided into blank control group(Ⅰ), model control group(Ⅱ), atorvastatin group(Ⅲ), puerarin group(Ⅳ) and combination group(Ⅴ), 10 in each group. Ⅰgroup did not be treated, and the other 4 groups used LPS combined with methylprednisolone to make the rat model of steroid-induced femoral head necrosis. At the same time of modeling, atorvastatin group, puerarin group and combination group were administered with atorvastatin, puerarin, and combination therapy once a day,respectively. After 8 consecutive weeks, the rats were sacrificed and samples were taken for examination. HE staining was used to investigate the femoral head necrosis in rats. The levels of serum BGP, OPN and TGF-β1 were detected by ELISA. The expressions of Bcl-2, Osx and Runx2 mRNA in femoral head were detected by RT-PCR. The contents of VEGF and p53 protein in femoral head were detected by WB. Immunohistochemistry was used to detect the content of Smo and Gli protein in the femoral head tissue and TUNNEL was used to detect apoptosis. Results Compared with the single drug, the combination group could significantly increase serum BGP, TGF-β1 content, osteosarcoma Osx, Runx2 mRNA expression and VEGF, p53 protein content (P<0.05).The serum OPN content, Bcl-2 mRNA expression and Smo and Gli protein content in femoral head tissues were significantly decreased (P<0.01), and the femoral head necrosis and apoptosis were improved. Conclusion Puerarin combined with atorvastatin can affect Hedgehog signaling pathway and reduce the degree of necrosis of the femoral head. |
