白藜芦醇对人视网膜色素上皮细胞氧化损伤的保护作用及机制研究

摘要:
目的观察白藜芦醇对过氧化氢诱导的人视网膜色素上皮细胞氧化损伤的保护作用。方法培养的人RPE细胞系分为正常对照组、过氧化氢损伤组和不同浓度白藜芦醇保护组,正常对照组不作任何处理,过氧化氢损伤组加入200mg/L过氧化氢,白藜芦醇保护组加入不同浓度(12.5mg/L、25mg/L、50mg/L、100mg/L)后加入200μmol/L过氧化氢。采用CCK-8检测细胞抑制率,SOD及MDA试剂盒检测细胞培养液中SOD与MDA含量。结果通过CCK-8法检测发现50mg/L、100mg/L白藜芦醇保护组作用2h,8h,24h可以明显降低细胞抑制率,且与氧化损伤组相比差异有统计学意义(P<0.05);白藜芦醇保护组50mg/L、100mg/L SOD活性与氧化损伤组相比明显升高,差异有统计学意义(P<0.05);白藜芦醇保护组12.5mg/L、25mg/L、50mg/L、100mg/L 2h、8h、24h MDA含量与氧化损伤组相比明显降低,差异有统计学意义(P<0.01)。结论白藜芦醇对过氧化氢诱导的人RPE细胞氧化损伤有一定的保护作用,提示白藜芦醇对AMD的预防和治疗有一定的作用。

Study on Protective Effect and Mechanism of Resveratrol on Human Retinal Pigment Epithelial Cells Injured by Hydrogen Peroxide

Abstract:
Objective To observe the protective effect of resveratrol on injury induced by hydrogen peroxide oxidation of human retinal pigment epithelial cells.Methods The cultured human RPE cell line was divided into normal control group,hydrogen peroxide damage group and different concentration resveratrol protective group.The normal control group without any treatment,hydrogen peroxide injury group with 200mg/L hydrogen peroxide,while the resveratrol protection group was added different concentrations( 12. 5mg/L 、25mg/L、50mg/L、100mg/L) and then was added 200μmol/L hydrogen peroxide.The cell inhibition rate was detected by CCK-8,and the contents of SOD and MDA in the cell culture medium were detected by SOD and MDA kits.Results The detectionby CCK-8 method showed that 50、100 mg/L of resveratrol for 2h,8h and 24 h could significantly reduce the rate of cell inhibition,and compared with the oxidized injury group,the difference was statistically significant( P < 0. 05). The activity of50mg/L、100mg/L of SOD in resveratrol protection groupwas significantly higher than that in the oxidative injury group,and the difference was statistically significant( P<0. 05).The contents of 12. 5mg/L、25mg/L、50mg/L、100mg/L of MDA at 2h,8h and 24 h in resveratrol protective group were significantly lower than those in oxidized injury group,and the difference was statistically significant( P<0. 01).Conclusion Resveratrol has protective effect on oxidative damage of RPE cells induced by hydrogen peroxide,suggesting a role in the prevention and treatment of resveratrol on AMD.
