目的探讨AFB1对人脐静脉内皮细胞(HUVEC)的毒性损伤及作用机制。方法不同浓度AFB1与HUVEC细胞共孵育后,采用CCK-8法检测细胞存活率,测定细胞LDH释放率,采用Annexin V/PI双染、DAPI染色检测细胞死亡机制。结果 AFB1对HUVEC细胞具有显著毒性作用,随着AFB1作用时间和剂量的增加,细胞存活率降低、LDH释放率增高;Annexin V/PI双染检测到细胞膜磷脂酰丝氨酸外翻;DAPI染色检测到细胞核断裂。结论 AFB1对HUVEC细胞有显著毒性损伤作用,其可能通过凋亡的机制引起细胞死亡。 |

Objective To investigate the toxic injury and mechanism of AFB1 on human umbilical vein endothelial cells( HUVEC).Methods After AFB1 and HUVEC cells were incubated at different concentrations,the survival rate of cells was detected by CCK-8,the release rate of LDH was determined,and the mechanism of cell death was detected by Annexin V/PI double staining and DAPI staining.Results AFB1 had significant toxic effects on HUVEC cells. With the increase of time and dose of AFB1,cell viability decreased and LDH release rate increased. Phosphatidylserine turnover in the cell membrane was detected by Annexin V/PI double staining.Nuclear fragmentation was detected by DAPI staining.Conclusion AFB1 has significant cytotoxicity in HUVEC cells,which may be through the mechanism of apoptosis induced cell death. |
