不同病变阶段OA软骨细胞中TGF-β信号通路相关分子的表达规律

摘要:
目的探讨OA软骨细胞中TGF-β信号通路相关分子(ALK1、ALK5、Smad3)及MMP13、Ⅱ型胶原的表达规律。方法将全膝关节置换或创伤性截肢膝关节OA软骨细胞,按病变阶段分组为早、中、晚期,通过Western-blot测定不同病变阶段的OA软骨细胞中ALK1、ALK5、Smad3及MMP13、Ⅱ型胶原的表达规律。结果 (1)OA软骨细胞随着病变程度的进展,ALK1、ALK5、Smad3及MMP13、Ⅱ型胶原在蛋白水平的表达量的变化差异有统计学意义(P<0.05),其中ALK1、ALK5、Smad3及Ⅱ型胶原随着病变程度的进展而出现下降,MMP13随着病变程度的进展出现上升;(2)OA软骨细胞随着病变的进展,ALK5的下降幅度大于ALK1,ALK1/ALK5的比例会随着病变程度的进展而出现上升。结论 OA软骨细胞中,TGF-β信号通路相关分子及其下游分子的蛋白表达量随着病变的增加而变化:ALK1/ALK5的比例及MMP13会随着病变程度的进展而出现上升,而ALK1、ALK5、Smad3、Ⅱ型胶原随着病变程度的进展而出现下降,并且ALK5下降的幅度大于ALK1。

Expression Law of TGF-β signaling Pathway Related Molecules in OA Chondrocytes at Different Stages of Lesion

Abstract:
Objective To investigate the expression pattern of TGF-βsignaling pathway related molecules(ALK1、ALK5、Smad3) and MMP13,type II collagen,in OA chondrocytes.Methods The OA chondrocytes of total knee replacement or traumatic amputation were divided into early,middle and late stages according to the stage of lesion. The expressions law of ALK1,ALK5,Smad3 and MMP13,type II collagen in OA chondrocytes at different pathological stages were determined by Western-blot. Results(1) The changes of the expression amountof ALK1,ALK5,Smad3 and MMP13,type II collagen in the protein level of OA chondrocytes were statistically significant with the progression of lesions. Among them,ALK1,ALK5,Smad3 and type II collagen decreased with the progression of the lesion,and MMP13 increased as the lesion progressed.(2) With the progression of the lesion,the decrease of ALK5 in OA chondrocytes was greater than ALK1,and the proportion of ALK1/ALK5 increased with the progression of lesion.Conclusion In OA chondrocytes,the expression of TGF-βsignaling pathway related molecules and their downstream molecules changed with the increase of leision: The proportion of ALK1/ALK5 and MMP13 increased with the progression of lesion,while ALK1,ALK5,Smad3 and type II collagen decreased with the progression of lesion,and the magnitude of ALK5 decrease was more than ALK1.
