糖尿病肾病大鼠足细胞转分化特征研究

摘要:
目的观察糖尿病肾病(diabetic nephropathy,DN)大鼠足细胞上皮细胞及间充质细胞标志物的变化,揭示DN大鼠足细胞转分化特征。方法分别应用免疫组织化学、实时定量PCR检测DN大鼠足细胞WT1、P-Cadherin、Desmin的阳性面积比、积分光密度及mRNA的表达,并与正常大鼠作对照;同时将DN组大鼠24h尿蛋白定量与上述各指标积分光密度进行相关性分析。结果与正常大鼠比较,DN组大鼠WT1下降,但差异无统计学意义(P>0.05);P-Cadherin降低,差异有统计学意义(P<0.05);而Desmin增高,差异有统计学意义(P<0.05);24h尿蛋白定量与WT1积分光密度无相关性(r=-0.337,P>0.05),与P-Cadherin积分光密度呈负相关(r=-0.600,P<0.05),与Desmin积分光密度呈正相关(r=0.750,P<0.01)。结论 DN大鼠早期(12周)足细胞数目尚未减少时,可能已经发生了上皮-间充质细胞转分化,而尿蛋白的发生与此有一定的相关性。

Characteristics of Podocyte Transdifferentiation in Diabetic Nephropathy Rats

Abstract:
Objective To observe the changes of markers of podocyte epithelial cells and mesenchymal cell in diabetic nephropathy( DN) rats,and to reveal the characteristics of podocyte transdifferentiation in DN rats. Methods WT1,P-Cadherin,positive area ratio of Desmin,integral optical density and expression of mRNAin podocytes of DN rats were detected by immunohistochemistry and real-time quantitative PCR,and they were compared with normal rats; At the same time,the analysis of correlation between 24 h urinary protein of rats in group DN and the integral optical density of above indexes was performed. Results Compared with the normal rats,the WT1 of rats in group DN decreased,but the difference was not statistically significant( P>0. 05);the P-Cadherin decreased,and the difference was statistically significant( P < 0. 05); however,the Desmin increased,and the difference was statistically significant( P<0. 05); there was no correlation between 24 h urinary protein quantification and WT1 integral optical density( r =-0. 337,P>0. 05),negatively correlated with integral optical density of P-Cadherin( r =-0. 600,P<0. 05),and positively correlated with integral optical density of Desmin( r = 0. 750,P<0. 01).Conclusion In the early stage( 12 weeks),when the number of podocytes in DN rats has not decreased,the epithelial mesenchymal transition may have occurred,and the occurrence of urinary proteins is related to this.
